Regular Transformation Protocol

  1. Thaw comp cell on ice (20 minutes).  DO NOT VORTEX, mix by gently flicking the tube.
  2. Distribute comp cells, 50 µl per tube into tubes on ice.
  3. Add 1 µl plasmid mixture, gently "flick" tube, and incubate on ice for 20 minutes.
  4. Heat shock for 45 seconds at 42 °C (if using microfuge tubes)
  5. Return to ice for 2 minutes.
  6. Add 75 µl of pre-warmed SOC medium and incubate at 30 °C with shaking for 60 minutes .
  7. Plate all of the mix on pre-warmed plates plus appropriate antibiotic.
